Description

Assay Genie offers a Cell-Based ELISA kit for the qualitative detection of cyclin-dependent kinase 10 (CDK10) in cultured cells. The assay uses an indirect ELISA format in which CDK10 is captured by target-specific primary antibodies and detected using HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition, read at 450 nm. The reported dynamic range is > 5000 cells.

The kit is designed for use with adherent cells and can be applied to cell lysates and culture supernatants. It allows researchers to measure relative levels of CDK10 expression and to screen for the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on CDK10. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalization, and Crystal Violet whole-cell staining may be used to determine cell density and adjust for plating differences. Reactivity is reported for human, mouse, and rat samples. Storage is at 4°C for 6 months.

CDK10 is described as a CMGC kinase of the CDK family with a role in cellular proliferation, functioning in the G2-M phase of the cell cycle. Biological processes associated with the kinase include negative regulation of cell proliferation, traversing the start control point of the mitotic cell cycle, positive regulation of the MAPKKK cascade, and protein amino acid phosphorylation. Dysregulation of CDK10 has been linked to various diseases, including cancer.
