Description

Built for accurate detection and quantification of CDC37 phosphorylated at Serine 13 in cell lysates and tissue extracts, Assay Genie's colorimetric cell-based ELISA kit provides high sensitivity and specificity for a variety of research applications. The assay uses an indirect ELISA format with colorimetric detection at 450 nm, in which HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition. It is reactive with Human, Mouse, and Rat samples and has a dynamic range of > 5000 Cells.

Designed as a qualitative, lysate-free, high-throughput assay, the kit enables measurement of relative amounts of phosphorylated CDC37 in cultured cells and can be used to screen the effects of various treatments, inhibitors, or activators on CDC37 phosphorylation. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining method may be used to determine cell density. The sample type is adherent cells.

CDC37 is a molecular chaperone involved in cell signal transduction, forming complexes with Hsp90 and a variety of protein kinases including CDK4, CDK6, SRC, RAF-1, MOK, and eIF2 alpha kinases, and interacting with the androgen receptor. It plays a critical role in directing Hsp90 to its target kinases. Phosphorylation of CDC37 at Serine 13 is important for its function in the cell cycle and cellular signaling pathways, and dysregulation of this phosphorylation has been implicated in cancer and neurodegenerative disorders. The protein is identified by Gene ID 11140 and UniProt ID Q16543, with a molecular weight of 378.
