Description

This colorimetric, cell-based ELISA kit from Assay Genie is designed for the qualitative detection of CDC25A phosphorylated at serine 178 in cultured cells, offering a lysate-free and high-throughput workflow. The assay uses an indirect ELISA format: phospho-specific primary antibodies capture the target and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition, with absorbance read at 450 nm. Reactivity is established for human, mouse and rat samples, and the dynamic range is > 5000 Cells.

The procedure is suited to adherent cells and can be used to measure relative levels of phosphorylated CDC25A in cultured cells as well as to screen treatments, inhibitors (e.g., siRNA or chemicals) or activators for their effects on CDC25A phosphorylation. A monoclonal anti-GAPDH antibody is included to serve as an internal positive control for normalizing target absorbance, and a Crystal Violet whole-cell staining method may be used to determine cell density. CDC25A is a dual-specificity phosphatase required for progression from G1 to the S phase of the cell cycle, where it activates cyclin-dependent kinase CDC2 by removing two phosphate groups; it is specifically degraded in response to DNA damage and has been described as an oncogene, making the kit relevant to cell cycle, cancer biology and related research areas.
