Description

Designed for the detection of phosphorylated CDC25A at serine 124, this Colorimetric Cell-Based ELISA Kit from Assay Genie uses an indirect ELISA format. The assay is cell-based and suitable for adherent cells, allowing researchers to measure relative phospho-CDC25A levels in cultured cells without preparing lysates. It can also be used to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on CDC25A phosphorylation.

The kit is reactive with Human, Mouse and Rat samples, and uses a colorimetric method read at 450 nm, with a stated dynamic range of > 5000 Cells. Detection relies on primary antibodies specific for CDC25A (Phospho-Ser124) paired with HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control in normalizing the target absorbance values, and Crystal Violet whole-cell staining may be used to determine cell density and adjust for plating differences.

CDC25A is a member of the MPI phosphatase family and is required for progression from G1 to the S phase of the cell cycle. It activates the cyclin-dependent kinase CDC2 by removing two phosphate groups and is specifically degraded in response to DNA damage. Phosphorylation at serine 124 is central to its activity and stability, making the kit relevant to research into cell cycle regulation and the role of CDC25A in diseases including cancer and neurodegenerative disorders.
