Description

A colorimetric cell-based ELISA kit from Assay Genie for the detection and quantification of CDC2 phosphorylated at Threonine 161 in cell samples. The assay uses an indirect ELISA format, with target-specific primary antibodies capturing the analyte and HRP-conjugated secondary antibodies enabling colorimetric readout at 450 nm upon substrate addition.

Designed as a lysate-free, high-throughput assay, the kit is reactive with human, mouse, and rat samples and has a dynamic range of > 5000 Cells. It can be used to measure relative amounts of phosphorylated CDC2 in cultured cells and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on CDC2 phosphorylation. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining method may be used to determine cell density following the colorimetric measurement.

The target is encoded by the CDK1 gene (Gene ID: 983, UniProt ID: P06493), a member of the Ser/Thr protein kinase family and a catalytic subunit of the M-phase promoting factor (MPF) that is essential for G1/S and G2/M phase transitions of the cell cycle. The kit supports research applications in cell biology, cancer research, and drug development.
