Description

From Assay Genie, this colorimetric, cell-based ELISA kit enables qualitative detection of phosphorylated CDC16/APC6 at serine 560 in cultured cells. It is reactive with human and mouse samples and is suited to research into cell cycle regulation, mitosis, and the control of cell division and chromosome segregation.

Detection uses an indirect ELISA format in which HRP-conjugated secondary antibodies bind CDC16/APC6 (Phospho-Ser560)-specific primary antibodies, with a colorimetric readout at 450 nm. The assay is lysate-free and high-throughput, allowing screening of various treatments, inhibitors (such as siRNA or chemicals), or activators on CDC16/APC6 phosphorylation. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance, and Crystal Violet whole-cell staining can be used to determine cell density and adjust for plating differences. The published dynamic range is > 5000 Cells.

CDC16/APC6 is a component of the anaphase-promoting complex/cyclosome (APC/C), a cell cycle-regulated E3 ubiquitin ligase that controls progression through mitosis and the G1 phase of the cell cycle. Phosphorylation at serine 560 has been implicated in the control of cell division and chromosome segregation, making the kit a useful tool for cell biology and cancer biology research.
