Description

A colorimetric cell-based ELISA kit from Assay Genie designed to detect CD5 protein expression in cultured adherent cells using a qualitative, lysate-free indirect ELISA format. The assay captures CD5 with target-specific primary antibodies while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction on substrate addition. Reported reactivity covers Human, Mouse, and Rat samples, with absorbance read at 450 nm, and the kit supports screening for the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on CD5 levels.

The kit is supplied in a 96-well microplate format with a reported dynamic range of > 5000 Cells and storage stability of 6 months at 4°C. Two normalization methods are included: a monoclonal antibody specific for human GAPDH serving as an internal positive control, and a Crystal Violet whole-cell staining approach to determine cell density, allowing absorbance values to be normalized to cell amounts and plating differences to be adjusted.

UniProt annotation (P06127) describes CD5 as a surface glycoprotein expressed on T cells and B-1 B cells. It is rapidly recruited to the immunological synapse, where it lowers the response of the T cell antigen receptor, interacts with CD72/LYB-2, and promotes B-cell survival through stimulation of autocrine IL-10 production. The protein contains 3 SRCR domains, has a reported molecular weight of 459, and is located at chromosomal position 11q13 in humans.
