CD40 (Phospho-Thr254)Colorimetric Cell-Based ELISA Kit

From Assay Genie comes a cell-based ELISA designed to detect phosphorylation of CD40 at threonine 254 in cultured cells. The assay uses a colorimetric readout at 450 nm in an indirect ELISA format, in which target-specific primary antibodies are bound by HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition.

It is a lysate-free, high-throughput, and sensitive kit intended for adherent cell samples, with reactivity to human and mouse. The stated dynamic range is greater than 5000 cells. Because the readout is qualitative, two normalization approaches are provided: a monoclonal anti-GAPDH antibody serves as an internal positive control, and Crystal Violet whole-cell staining may be used to determine cell density and adjust for plating differences.

CD40 is a member of the TNF-receptor superfamily and a cell surface receptor involved in immune and inflammatory signalling, with phosphorylation at Thr254 reported to play a role in downstream pathways. Dysregulation of CD40 signalling has been implicated in autoimmune disorders and cancer, making this phospho-specific assay relevant to research in immunology, oncology, and related fields.
