Description

This Cell-Based ELISA Kit from Assay Genie is designed for the accurate detection of phosphorylation at tyrosine 292 of the CD32 protein in cell lysates and tissue samples. Using a colorimetric method measured at 450 nm, the assay operates via an indirect ELISA format in which phospho-specific primary antibodies capture the target and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition. The system is described as lysate-free, high throughput, and sensitive, intended for measuring relative amounts of phosphorylated CD32 in cultured cells and for screening the effects of various treatments, inhibitors, or activators on CD32 phosphorylation.

Reactivity is reported for Human, Mouse, and Rat samples, with a dynamic range of > 5000 Cells. Two normalization approaches are included: a monoclonal antibody specific for human GAPDH serves as an internal positive control in normalizing the target absorbance values, and a Crystal Violet whole-cell staining method may be used to determine cell density after the colorimetric measurement, allowing absorbance values to be normalized to cell amounts. The sample type is adherent cells, and the kit is positioned within the Immunology research area.

CD32, also known as FcγRII, is a cell surface receptor involved in immune responses and inflammation, with phosphorylation at tyrosine 292 playing a role in modulating receptor function and downstream signaling pathways. Dysregulation of CD32 phosphorylation has been implicated in autoimmune disorders and cancer, making it a valuable biomarker for studying these conditions and developing targeted therapies. The associated gene is FCGR2B (Gene ID: 2213, UniProt ID: P31994).
