Description

From Assay Genie, this Colorimetric Cell-Based ELISA Kit provides a cell-based, lysate-free, high-throughput method for the detection of CD3 zeta protein levels in cultured cells. The assay uses an indirect ELISA format, in which CD3 zeta-specific primary antibodies capture the target and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, producing a colorimetric reaction upon substrate addition that is read at 450 nm.

Reactivity is listed for Human, Mouse, and Rat samples, with a dynamic range of > 5000 Cells. The kit is supplied as a 96-Well Microplate format, intended for storage at 4°C with a stated stability of 6 months. To support data normalisation, a monoclonal antibody specific for human GAPDH is included as an internal positive control, and a Crystal Violet whole-cell staining step may be used to determine cell density and adjust for plating differences. Adherent cells are supported in the standard procedure.

CD3 zeta, encoded by the CD247 gene, is a T-cell surface glycoprotein that forms part of the T-cell antigen receptor complex, coupling antigen recognition to intracellular signal-transduction pathways. Dysregulation of CD3 zeta has been associated with autoimmune diseases, immunodeficiencies, and cancer, making it a relevant target in T-cell biology, immune signalling, and immunology research. The kit can be applied to measure relative CD3 zeta amounts in cultured cells and to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on CD3 zeta expression.
