Description

A cell-based, colorimetric ELISA kit from Assay Genie designed for the qualitative detection of CD253 protein expression directly in cultured cells. The indirect ELISA format uses CD253-specific primary antibodies for capture, with HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition, measured at 450 nm. The assay is described as lysate-free and high-throughput, suited for measuring relative CD253 levels and for screening the effects of treatments, inhibitors such as siRNA or chemicals, or activators on CD253 expression.

The kit is configured for human samples with a stated dynamic range of > 5000 cells and a 96-well microplate format. It includes a monoclonal anti-human GAPDH antibody to serve as an internal positive control for normalizing target absorbance, and a Crystal Violet whole-cell staining step for determining cell density so absorbance values can be normalized to cell numbers. CD253, also known as TRAIL (TNF-related apoptosis-inducing ligand), is a cytokine of the TNF ligand family involved in the regulation of apoptosis, with roles in the immune response and tumor suppression, and has been implicated in cancer and autoimmune disorders.

Sample types cited for detection include cell lysates, serum, plasma, and tissue homogenates, and the product is also indicated for adherent cells. The assay is positioned for research in oncology, immunology, and cell biology, including studies of cell death. Storage is at 4°C with a stated stability of 6 months.
