Description

A colorimetric cell-based ELISA from Assay Genie enables qualitative measurement of CD226 protein expression directly in cultured cells. The assay uses an indirect ELISA format in which CD226 is captured by target-specific primary antibodies, while HRP-conjugated secondary antibodies binding the Fc region of the primary antibody catalyze a colorimetric reaction upon substrate addition. Designed as a lysate-free, high-throughput workflow, the kit can also be applied to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on CD226.

The kit is reactive with human and mouse samples, with a dynamic range of > 5000 Cells and detection by colorimetric measurement at 450 nm. It is supplied in a 96-Well Microplate format with storage at 4°C for 6 Months. Because the assay is qualitative, two normalization approaches are supported: a monoclonal antibody specific for human GAPDH serves as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining method may be used to determine cell density so that absorbance values can be normalized to cell amounts.

CD226, also known as DNAM-1, is a key protein involved in regulating immune responses, including natural killer cell activation and T cell-mediated cytotoxicity. Dysregulation of CD226 has been implicated in autoimmune disorders and cancer, supporting its value as a potential biomarker for disease diagnosis and treatment monitoring. The kit is intended for research in cell biology and immunology.
