Description

From Assay Genie, this cell-based ELISA kit is designed for the precise measurement of CD19 phosphorylation at tyrosine 531 in cultured cells. The kit offers a convenient, lysate-free, high-throughput and sensitive workflow that detects the CD19 protein phosphorylation and expression profile in cells. It can be used to measure the relative amounts of phosphorylated CD19 in cultured cells as well as to screen for the effects that various treatments, inhibitors (such as siRNA or chemicals), or activators have on CD19 phosphorylation.

Qualitative determination of CD19 (Phospho-Tyr531) concentration is achieved by an indirect ELISA format. The target is captured by CD19 (Phospho-Tyr531)-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, allowing the HRP enzyme to catalyse a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and the Crystal Violet whole-cell staining method may be used to determine cell density. The assay is reactive with human and mouse samples and is intended for use with adherent cells, with a dynamic range of > 5000 Cells and detection at 450 nm.

CD19 is a key signalling molecule in B-cell activation and function, and assembles with the antigen receptor of B-lymphocytes to decrease the threshold for antigen receptor-dependent stimulation. Phosphorylation of CD19 at tyrosine 531 is a crucial event in regulating B-cell signalling pathways, making this kit a useful tool for research into immune-related disorders, autoimmune diseases and novel drug targets.
