Description

Assay Genie offers a cell-based ELISA kit for the qualitative detection of CD147 protein expression directly in cultured cells. The immunoassay uses an indirect format in which CD147 is captured by target-specific primary antibodies and detected with HRP-conjugated secondary antibodies that catalyze a colorimetric reaction, read at 450 nm. The kit is described as lysate-free, high-throughput and sensitive, and is intended for use with adherent cells.

The kit is reactive with Human, Mouse and Rat samples and has a stated dynamic range of > 5000 Cells. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining step can be used to determine cell density so that absorbance values can be normalized to cell number. CD147, also known as EMMPRIN or basigin (gene symbol BSG), is a transmembrane glycoprotein implicated in tumor invasion, inflammation and immune responses, with overexpression linked to cancer progression and metastasis.

Storage is at 4°C with a stated stability of 6 Months. The procedure follows a standard cell-based ELISA workflow: cells are seeded, fixed, quenched, blocked, incubated overnight with primary antibodies, incubated with HRP-conjugated secondary antibodies, developed with ready-to-use substrate and read at 450 nm after addition of stop solution.
