CCR5 (Phospho-Ser349)Colorimetric Cell-Based ELISA Kit

From Assay Genie, this Cell-Based ELISA Kit is designed to detect phosphorylation of CCR5 at Ser349 in cultured cells. It uses an indirect ELISA format, in which a phospho-specific primary antibody captures the target and an HRP-conjugated secondary antibody catalyses a colorimetric reaction read at 450 nm. The assay is lysate-free, with a stated dynamic range of > 5000 cells, and is reported for use with human samples. The kit can also be applied to screen the effects of various treatments, inhibitors or activators on CCR5 phosphorylation.

A monoclonal anti-GAPDH antibody is included to serve as an internal positive control for normalising target absorbance values. Because the Cell-Based ELISA is qualitative, an additional normalisation step is recommended: following the HRP readout, a Crystal Violet whole-cell stain can be used to determine cell density, allowing absorbance values to be normalised to cell amount and adjust for plating differences.

CCR5 is a 7-transmembrane G-protein-coupled receptor for inflammatory C-C chemokines including MIP-1-alpha, MIP-1-beta and RANTES, and acts as a co-receptor (alongside CD4) for HIV-1 R5 isolates. The corresponding human protein has UniProt ID P51681 and the gene maps to chromosomal location 3p21.31.
