Description
This in-cell ELISA from Assay Genie is designed for the qualitative detection of C-C chemokine receptor type 5 (CCR5) in cultured human cells. Using a colorimetric readout at 450 nm in a 96-well microplate format, the assay measures relative CCR5 protein expression in adherent cell populations. The procedure is lysate-free and high-throughput, allowing researchers to screen treatments, inhibitors, or activators that may modulate CCR5 levels.
The kit employs an indirect ELISA format: CCR5 is captured by target-specific primary antibodies, which are then bound by HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. Because the readout is qualitative, two normalization strategies are included. A monoclonal anti-human GAPDH antibody serves as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining step allows normalization of absorbance readings to cell density.
CCR5 is a seven-transmembrane G-protein-coupled receptor for inflammatory C-C chemokines such as MIP-1α, MIP-1β and RANTES, and functions as a co-receptor (along with CD4) for HIV-1 R5 isolates. Storage is at 4°C with a stated stability of 6 months.
