Description

This Cell-Based ELISA kit from Assay Genie enables accurate detection of phospho-Tyr654 levels of β-catenin in cell lysates. The assay uses an indirect ELISA format in which Catenin-beta (Phospho-Tyr654) is captured by phospho-specific primary antibodies, with HRP-conjugated secondary antibodies binding the Fc region of the primary antibody. Substrate addition then drives a colorimetric reaction read at 450 nm.

The kit is described as lysate-free, high throughput, and sensitive, and can be used to measure relative amounts of phosphorylated Catenin-beta in cultured cells. It is also suitable for screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on Catenin-beta phosphorylation. Reported reactivity covers Human, Mouse, and Rat samples, and the assay's dynamic range supports analysis of more than 5000 cells. Sample type is given as adherent cells.

For data normalization, a monoclonal antibody specific for human GAPDH is included as an internal positive control. Following the colorimetric HRP measurement, Crystal Violet whole-cell staining may be used to determine cell density, with results then normalized to cell amounts to adjust for plating differences. The target corresponds to the CTNNB1 gene (Gene ID: 1499; UniProt ID: P35222), a key downstream effector in the Wnt signaling pathway that regulates cell proliferation, differentiation, and migration, with phosphorylation at Tyr654 being critical for its stability and function.
