Description

From Assay Genie, this Cell-Based ELISA Kit is designed for the qualitative measurement of Catenin-beta (Phospho-Tyr489) phosphorylation in cultured adherent cells. The assay uses a colorimetric detection method measured at 450 nm and has been validated for reactivity with Human, Mouse, and Rat samples, with a dynamic range of > 5000 Cells. The kit is provided in a 96-Well Microplate format and is described as lysate-free, high throughput, and sensitive.

Detection is achieved by an indirect ELISA format in which phospho-Tyr489 Catenin-beta is captured by target-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values. Following the colorimetric measurement of HRP activity, the Crystal Violet whole-cell staining method may be used to determine cell density, allowing absorbance values to be normalized to cell amounts.

The target, Catenin-beta (gene symbol CTNNB1; Gene ID 1499; UniProt P35222), is a regulator of cell adhesion and a key downstream effector in the Wnt signaling pathway, with roles in cell proliferation, differentiation, and migration. Phosphorylation at tyrosine 489 has been shown to regulate Catenin-beta activity and stability, influencing downstream signaling relevant to cancer, development, and tissue regeneration. The kit supports screening for the effects of treatments, inhibitors, and activators on Catenin-beta phosphorylation.
