Description

From Assay Genie, the Catenin-beta (Phospho-Ser37) Colorimetric Cell-Based ELISA Kit is designed for the accurate and reliable detection of phosphorylated beta-Catenin at Ser37 in cells. The kit is reactive with Human, Mouse and Rat samples and is intended for use with adherent cells. Detection is colorimetric, measured at 450 nm, with a dynamic range of > 5000 Cells. Because beta-Catenin is a key player in the Wnt signaling pathway, where it influences gene expression and cell differentiation, phosphorylation at Ser37 is an important research target, with implications in diseases including cancer and developmental disorders.

The Cell-Based ELISA is lysate-free, high throughput and sensitive, and can be used to measure the relative amounts of phosphorylated Catenin-beta in cultured cells as well as to screen for the effects that various treatments, inhibitors or activators have on Catenin-beta phosphorylation. The assay uses an indirect format, in which Catenin-beta (Phospho-Ser37)-specific primary antibodies are bound by HRP-conjugated secondary antibodies, allowing a colorimetric reaction to be generated upon substrate addition. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining method is provided for determining cell density so that absorbance readings can be adjusted for plating differences.
