Description

A cell-based ELISA kit from Assay Genie designed for the qualitative detection of Caspase 8 phosphorylated at Tyr380 in cultured human cells. The assay uses an indirect ELISA format in which target phosphorylation is captured by phospho-specific primary antibodies and detected with HRP-conjugated secondary antibodies that catalyze a colorimetric reaction measured at 450 nm. A monoclonal GAPDH antibody is included as an internal positive control for normalization, and a Crystal Violet whole-cell staining step can be used to normalize absorbance values to cell density, allowing plating differences to be adjusted. The kit is lysate-free and intended for use with adherent cells, supporting high-throughput screening of treatments such as siRNA or chemical modulators of Caspase 8 phosphorylation.

According to the manufacturer's datasheet, the target is the CASP8 gene product (Gene ID: 841, UniProt ID: Q14790), and the kit has a stated dynamic range of > 5000 Cells. Caspase 8 is described as the most upstream protease of the caspase activation cascade responsible for FAS- and TNFRSF1A-induced cell death, with dysregulation of its phosphorylation implicated in cancer, autoimmune disorders, and neurodegenerative conditions.
