Description

Assay Genie's cell-based ELISA kit is designed for the detection of phosphorylated Caspase-8 at Ser347 in cell lysates. The kit is a lysate-free, high-throughput assay that uses an indirect ELISA format in which the target is captured by phospho-specific primary antibodies and detected using HRP-conjugated secondary antibodies that catalyze a colorimetric reaction at 450 nm upon substrate addition.

It can be used to measure relative amounts of Caspase-8 (Phospho-Ser347) in cultured cells and to screen for the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on Caspase-8 phosphorylation. The kit is reactive with human and rat samples, has a dynamic range of > 5000 Cells, and is intended for use with adherent cells. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining method may be used to determine cell density for additional normalization.

The target is Caspase-8 (gene symbol CASP8), an initiator protease of the caspase activation cascade responsible for death receptor-mediated apoptosis, with the phosphorylation site of interest being Ser347. The qualitative nature of the assay makes it suited for research into apoptotic signaling and cell death regulation.
