Caspase 3 Colorimetric Cell-Based ELISA Kit

This cell-based ELISA kit from Assay Genie is designed for the qualitative detection of Caspase 3 protein expression in cultured cells. Using an indirect ELISA format, the kit captures Caspase 3 with target-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody and catalyze a colorimetric reaction upon substrate addition. It is described as convenient, lysate-free, and suitable for high-throughput and sensitive workflows, and can be used to measure relative Caspase 3 amounts in cultured cells as well as to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on Caspase 3. Detection is colorimetric, and the assay is reactive with Human, Mouse, and Rat samples, with a stated dynamic range of > 5000 Cells. The recommended sample type is adherent cells, making the kit relevant for studies of apoptosis and caspase-3 biology in which dysregulation of caspase-3 activity has been implicated in cancer, neurodegenerative disorders, and autoimmune conditions.

Because the Cell-Based ELISA readout is qualitative, two normalization approaches are provided: a monoclonal antibody specific for human GAPDH serves as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining step may be used to determine cell density, allowing absorbance values to be adjusted to cell amounts. The kit is supplied in a microplate format, with recommended storage at 4°C and a stated stability of 6 Months.
