Description

This Cell-Based ELISA Kit from Assay Genie is designed for the qualitative determination of Casein Kinase I alpha phosphorylated at the Tyr294 site in adherent cultured cells. The kit uses an indirect ELISA format in which a phospho-specific primary antibody captures the target, and an HRP-conjugated secondary antibody catalyses a colorimetric reaction read at 450 nm.

Reactivity has been validated against Human, Mouse, and Rat samples, with a stated dynamic range of > 5000 Cells. The assay is lysate-free and suitable for high-throughput screening of treatments, inhibitors (such as siRNA or chemicals), or activators that modulate Casein Kinase I alpha phosphorylation. A monoclonal anti-GAPDH antibody is included as an internal positive control, and a Crystal Violet whole-cell staining step allows normalisation of absorbance values to cell density.

Casein Kinase I alpha (gene symbol CSNK1A1) is a serine/threonine protein kinase of the CK1 family that participates in Wnt signalling by phosphorylating beta-catenin, marking it for ubiquitin-dependent proteasomal degradation. The kit is classified within the Cell Cycle research area as a Phospho sub-type.
