Description

From Assay Genie, this Sandwich ELISA kit provides quantitative measurement of TNF-Beta (Tumor Necrosis Factor Beta) in canine samples using a colorimetric detection method with a double antibody configuration. The assay delivers a sensitivity of 9.375 pg/mL across a working range of 15.625-1000 pg/mL. Compatible sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, with all components intended for storage at 2-8°C for 12 months.

Validated linearity spans 82-102% across serum, EDTA plasma, and heparin plasma at dilutions of 1:2, 1:4, and 1:8 (n=5). Recovery performance is similarly consistent, with serum averaging 96% (range 93-102%), EDTA plasma averaging 95% (range 88-99%), and heparin plasma averaging 95% (range 89-101%). The protocol follows a sequential workflow: reagent and plate preparation, primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin (SABC) binding at 37°C for 30 minutes, TMB substrate color development in the dark for 10-20 minutes, and absorbance measurement at 450 nm after adding stop solution. Sample-specific preparation guidance covers serum, plasma, tissue homogenate, cell culture supernatant, and cell lysate processing.

TNF-Beta is a cytokine involved in inflammatory processes, immune response modulation, and cell apoptosis, with dysregulation implicated in autoimmune disorders, infectious diseases, and cancer. The kit supports research into TNF-Beta biology and disease pathogenesis in canine models, with Uniprot reference Q5WR07.
