Description

From Assay Genie, a sandwich double-antibody ELISA enables quantitative measurement of canine MCP-1/CCL2 in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The microplate is pre-coated with a capture antibody specific to the target protein, and the workflow uses a biotinylated detection antibody followed by HRP-conjugated Streptavidin to generate a colorimetric reaction that is stopped and read at 450 nm. Related protein identifiers listed for the analyte include MCP-1, CCL2, GDCF-2, HC11, HSMCR30, MCAF, MCP1, SCYA2 and SMC-CF (UniProt P52203).

Sensitivity is reported at 9.375 pg/mL with a working range of 15.625–1000 pg/mL. Spike-and-recovery values across n=5 measurements are 98% average (range 89–103%) for serum, 93% average (range 89–101%) for EDTA plasma, and 96% average (range 85–105%) for heparin plasma. Linearity at 1:2, 1:4 and 1:8 dilutions is given as 86–103%, 86–99% and 85–97% for serum; 87–104%, 82–95% and 81–99% for EDTA plasma; and 95–103%, 84–101% and 83–100% for heparin plasma. Kits are stored at 2–8°C for 12 months.

The procedure proceeds through reagent and plate preparation, primary incubation at 37°C for 90 minutes, biotinylated detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin (SABC) binding at 37°C for 30 minutes, TMB substrate incubation in the dark for 10–20 minutes, and a stop step before absorbance is read at 450 nm.
