Description

From Assay Genie, this sandwich ELISA kit is designed for the quantitative measurement of interleukin-6 (IL-6) in canine samples. The assay uses a double-antibody sandwich format with colorimetric detection and is reactive with canine serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The reported sensitivity is 18.75 pg/mL and the standard curve range is 31.25–2000 pg/mL. IL-6 is catalogued under UniProt P41323 (KEGG K05405; Gene ID 403985) and is recognised by the synonyms IL6, BSF2, HGF, HSF, IFNB2, and CDF.

Supplied as a solid-phase ELISA, the kit is intended to be stored at 2–8°C for 12 months. Linearity was assessed at dilutions of 1:2, 1:4, and 1:8, with values of 85–102%, 87–97%, and 87–99% in serum; 87–99%, 83–95%, and 80–97% in EDTA plasma; and 87–105%, 84–99%, and 85–100% in heparin plasma. Spike-and-recovery testing returned averages of 96% in serum (range 87–102%), 97% in EDTA plasma (range 92–105%), and 91% in heparin plasma (range 86–97%).

The protocol consists of six stages: reagent and plate preparation; sample and standard loading with incubation at 37°C for 90 minutes; biotin-labeled detection antibody incubation at 37°C for 60 minutes; HRP-Streptavidin (SABC) incubation at 37°C for 30 minutes; TMB substrate development in the dark for 10–20 minutes; and stop solution addition followed by absorbance reading at 450 nm. Sample preparation guidance covers serum, plasma, tissue homogenate, cell culture supernatant, and cell lysate. Interleukin-6 is described as a key cytokine involved in immune response, inflammation, and the regulation of cellular processes.
