Manufactured by Assay Genie for canine research, this Sandwich ELISA uses a double-antibody, colorimetric format to quantify interleukin-10 (IL-10) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit achieves a sensitivity of 9.375 pg/mL across a standard range of 15.625-1000 pg/mL. IL-10 is an anti-inflammatory cytokine that regulates immune responses and helps maintain immune balance, and its dysregulation has been associated with autoimmune disorders, inflammatory conditions, and cancer.

Reagents are stored at 2-8°C for 12 months. Reported recovery ranges are 89-105% for serum (average 99%), 86-104% for EDTA plasma (average 97%), and 87-103% for heparin plasma (average 96%). The workflow includes equilibrating reagents to room temperature, loading standards and samples and incubating at 37°C for 90 minutes, adding a biotin-labeled detection antibody and incubating for 60 minutes, adding HRP-Streptavidin for 30 minutes, developing with TMB substrate in the dark for 10-20 minutes, then adding stop solution and reading absorbance at 450 nm. Serum is prepared by allowing blood to clot and centrifuging at 1000 × g for 20 minutes; plasma is collected using anticoagulant tubes and centrifuged at 1000 × g for 15 minutes at 2-8°C.
