Description

Assay Genie's Canine IFN gamma ELISA Kit is a sandwich-format immunoassay designed for the precise measurement of interferon-gamma (IFN-gamma) levels in canine samples. The kit uses a double-antibody sandwich ELISA configuration with colorimetric detection, and is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Interferon-gamma is a key cytokine involved in immune regulation, particularly in the activation of macrophages and the modulation of inflammatory processes, and is implicated in infectious diseases, autoimmune disorders, and cancer, making it a valuable biomarker for studying immune function and disease progression.

Reported performance characteristics include a sensitivity of 37.5 pg/mL and a standard curve range of 62.5-4000 pg/mL. Linearity across dilutions (1:2, 1:4, 1:8) has been demonstrated in serum (87-104%, 88-101%, 87-96%), EDTA plasma (86-104%, 83-99%, 81-97%) and heparin plasma (85-97%, 87-101%, 87-94%). Spike-and-recovery performance has been reported for serum (86-100%, average 95%), EDTA plasma (85-104%, average 96%) and heparin plasma (86-98%, average 93%). The kit is stored at 2-8°C and is stated to be stable for 12 months under those conditions. The associated UniProt entry is P42161.

The assay procedure involves equilibrating reagents and TMB substrate to room temperature; incubating standards, samples and blanks on the pre-coated plate at 37°C for 90 minutes; adding a biotin-labeled detection antibody with a 37°C incubation for 60 minutes; adding HRP-Streptavidin (SABC) with a 37°C incubation for 30 minutes; developing colour with TMB substrate in the dark for 10-20 minutes; and reading absorbance at 450 nm immediately after adding stop solution.
