Description

This Cell-Based ELISA Kit from Assay Genie is designed for detecting and quantifying phosphorylated CAMK2A and CAMK2D proteins at Thr286 in cell lysates. The kit uses a colorimetric detection method at 450 nm and has been validated for reactivity with Human, Mouse, and Rat samples, with a dynamic range of more than 5000 cells.

The assay employs an indirect ELISA format in which CAMK2A/CAMK2D (Phospho-Thr286) is captured by target-specific primary antibodies, with HRP-conjugated secondary antibodies binding the Fc region of the primary antibody. Upon substrate addition, the HRP enzyme catalyzes a colorimetric reaction, enabling qualitative determination of phosphorylated CAMK2A/CAMK2D. A monoclonal antibody specific for human GAPDH is included as an internal positive control to normalize target absorbance values. Following colorimetric measurement, the Crystal Violet whole-cell staining method may be used to determine cell density, allowing results to be normalized to cell amounts and adjusting for plating differences.

CAMK2A and CAMK2D are key proteins involved in cell signaling pathways, playing critical roles in processes such as learning and memory, muscle contraction, and neuronal plasticity. Phosphorylation at Thr286 is particularly important for their activation and regulation of various cellular functions, making this assay useful for investigating these proteins in health and disease, cellular signaling pathways, drug development, and disease mechanisms.
