Description

A cell-based ELISA from Assay Genie designed to measure CaMK1-alpha phosphorylation at threonine 177 and expression profiles in cultured cells. The kit uses a colorimetric detection method read at 450 nm, is reactive with human, mouse, and rat samples, and is suitable for adherent cells, with a dynamic range of > 5000 cells.

CaMK1 alpha is a calcium/calmodulin-dependent protein kinase involved in cellular processes such as cell growth, differentiation, and survival, and phosphorylation at Thr177 regulates its activity. The indirect ELISA format captures the target with phospho-specific or total primary antibodies, with HRP-conjugated secondary antibodies catalysing the colorimetric reaction. A monoclonal anti-GAPDH antibody is included as an internal positive control, and Crystal Violet staining can be used to normalise absorbance values to cell density. The assay can be used to screen the effects of treatments, inhibitors (including siRNA and chemicals), and activators on CaMK1-alpha phosphorylation. Gene Symbol: CAMK1 (Gene ID 8536, UniProt Q14012).

According to the manufacturer, phosphorylation of CAMK1 alpha at threonine 177 makes it a relevant target for studying signalling pathways and the mechanisms underlying diseases such as cancer, neurodegenerative disorders, and cardiovascular diseases.
