Description

A cell-based ELISA kit from Assay Genie is designed for the qualitative measurement of phosphorylated Calsenilin/KCNIP3 (Phospho-Ser63) in cultured cells. The assay uses an indirect ELISA format in which Calsenilin/KCNIP3 (Phospho-Ser63) is captured by a phospho-specific primary antibody, with HRP-conjugated secondary antibodies binding the Fc region of the primary antibody to catalyze a colorimetric reaction upon substrate addition.

The kit can be used to measure relative amounts of phosphorylated Calsenilin/KCNIP3 in cultured cells and to screen for the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on Calsenilin/KCNIP3 phosphorylation. It reacts with human and mouse samples and has a dynamic range of > 5000 Cells. Detection is colorimetric at 450 nm. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining step is provided to determine cell density for further normalization.

Calsenilin/KCNIP3 is a small calcium-binding protein belonging to the family of voltage-gated potassium (Kv) channel-interacting proteins (KCNIPs) within the recoverin branch of the EF-hand superfamily. It modulates Kv4 voltage-gated potassium channels and may regulate A-type currents and neuronal excitability in response to changes in intracellular calcium. It also functions as a calcium-regulated transcriptional repressor and interacts with presenilins, with dysregulation having been linked to neurodegenerative diseases such as Alzheimer's and Parkinson's.
