Description

From Assay Genie, this Colorimetric Cell-Based ELISA Kit is designed for the precise measurement of Calnexin phosphorylation at serine 583 in cell lysates. The kit uses a colorimetric detection method read at 450 nm and is reactive with Human, Mouse, and Rat samples. It employs a Cell-Based ELISA format with a dynamic range of > 5000 Cells, making it suitable for high-throughput screening of adherent cells.

The assay uses a lysate-free, indirect ELISA format in which Calnexin (Phospho-Ser583) is captured by specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. The HRP enzyme then catalyzes a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, and the Crystal Violet whole-cell staining method can be used to determine cell density for further normalization. The kit is intended for measuring the relative amounts of phosphorylated Calnexin in cultured cells, as well as for screening the effects of various treatments, inhibitors (e.g. siRNA or chemicals), or activators on Calnexin phosphorylation.

Calnexin is a calcium-binding, endoplasmic reticulum-associated protein of the calreticulin family that plays a key role in protein folding and quality control by interacting with newly synthesized N-linked glycoproteins. Phosphorylation at serine 583 has been linked to various cellular processes and signaling pathways, making this kit a valuable tool for studying cell signaling mechanisms and disease progression in immunology research.
