Calnexin Colorimetric Cell-Based ELISA Kit

This colorimetric cell-based ELISA kit from Assay Genie provides a lysate-free, high-throughput and sensitive method to detect and quantify calnexin protein in cultured cells. The qualitative assay can be used to measure relative calnexin levels in cells and to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on calnexin expression.

Detection follows an indirect ELISA format in which calnexin-specific primary antibodies capture the target, and HRP-conjugated secondary antibodies catalyze a colorimetric reaction read at 450 nm. The dynamic range is greater than 5000 cells, with reactivity against human, mouse, and rat samples. The kit is supplied in a 96-well microplate format suitable for adherent cells, with a storage stability of 4 °C for 6 months.

A monoclonal anti-GAPDH antibody is included as an internal positive control for normalization of target absorbance, and a Crystal Violet whole-cell staining step is provided to determine cell density and adjust for plating differences. Calnexin is encoded by the CANX gene (Gene ID 821; UniProt P27824) and is a calcium-binding, type I integral membrane protein of the endoplasmic reticulum that interacts with newly synthesized glycoproteins and contributes to the retention of incorrectly folded proteins as part of ER quality control.
