Description

This Colorimetric Cell-Based ELISA Kit from Assay Genie is designed for the detection of phosphorylated Calcium Sensing Receptor (CaSR) at Thr888 in cell samples. The phospho-specific assay provides a qualitative, lysate-free, high-throughput format suitable for measuring the relative amounts of phosphorylated CaSR in cultured cells, and for screening the effects of treatments, inhibitors, or activators on CaSR phosphorylation.

Reactivity has been established for Human, Mouse and Rat samples, with a dynamic range of > 5000 Cells. Detection is achieved colorimetrically at 450 nm using an indirect ELISA format, in which target-specific primary antibodies are captured and HRP-conjugated secondary antibodies catalyze the substrate reaction. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalization, and a Crystal Violet whole-cell staining step can be used to determine cell density.

CaSR is a G-protein coupled receptor that acts as a calcium sensor for parathyroid and kidney, helping to maintain a stable extracellular calcium concentration and regulating cellular functions including hormone secretion and cell proliferation. Phosphorylation at Thr888 is important for receptor activation and downstream intracellular signaling, making this kit a useful tool for research into calcium signaling and related disorders such as hypercalcemia and hypocalcemia.
