Description

This Cell-Based ELISA Kit from Assay Genie is designed to detect and quantify phosphorylation of C-RAF at tyrosine 341 in cultured cells. Using a lysate-free, indirect ELISA format with colorimetric detection at 450 nm, the kit enables researchers to measure relative levels of phosphorylated C-RAF and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on C-RAF phosphorylation.

The assay has been validated for Human, Mouse, and Rat samples with adherent cells, and supports a dynamic range of > 5000 Cells. Normalization is achieved through two methods: a monoclonal antibody specific for human GAPDH is included to serve as an internal positive control, and the Crystal Violet whole-cell staining method may be used to determine cell density, allowing absorbance values to be adjusted for plating differences.

C-RAF (also known as RAF1) is a key serine/threonine kinase in the MAPK/ERK signaling cascade, acting downstream of the Ras family of membrane-associated GTPases to regulate cell proliferation, differentiation, and survival. Phosphorylation at tyrosine 341 is known to regulate C-RAF activity and downstream signaling, making it a relevant target for research into cancer, inflammation, and other disease processes.
