Description

This cell-based ELISA kit from Assay Genie is designed for the detection of C-RAF phosphorylated at Ser338 in cultured adherent cells, providing a convenient, lysate-free, high-throughput method to study phosphorylation of this key kinase. Phosphorylation of c-Raf at serine 338 is implicated in the regulation of cell proliferation, differentiation and survival, and its dysregulation has been linked to diseases including cancer, making it a relevant target for signalling research and drug development.

The assay uses an indirect ELISA format in which C-RAF (Phospho-Ser338) is captured by phospho-specific primary antibodies and detected via HRP-conjugated secondary antibodies that catalyse a colorimetric reaction measured at 450 nm. The kit has a dynamic range of greater than 5000 cells and is reactive with human, mouse and rat samples. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalisation, and a Crystal Violet whole-cell staining step is provided to account for cell density differences across wells.

Qualitative determination of C-RAF (Phospho-Ser338) levels enables researchers to screen the effects of treatments, inhibitors (such as siRNA or chemicals) or activators on C-RAF phosphorylation. The target corresponds to the RAF1 gene (Gene ID 5894, UniProt ID P04049), a proto-oncogenic TKL kinase that links membrane-associated Ras GTPases to the MAPK/ERK cascade and influences cell fate decisions including proliferation, differentiation, apoptosis and survival.
