The c-Jun Transcription Factor Activity Assay from Assay Genie is a DNA-binding ELISA kit built on a microplate format, configured to capture and quantify activated c-Jun that has bound a consensus AP-1 response element. The plate is pre-coated with a double-stranded oligonucleotide containing the AP-1 recognition sequence, so only transcription factor complexes that recognize this site are retained. Nuclear and cell lysates prepared from human, mouse, and rat sources can be analyzed in this assay. The kit is supplied complete with coated microplate wells, primary anti-c-Jun antibody, HRP-conjugated secondary antibody, TMB substrate, and all necessary assay and wash buffers.

This assay is commonly used to monitor AP-1 transcriptional activity downstream of MAPK and JNK signaling, including phosphorylation of c-Jun at Ser63 and Ser73, and to compare activation states between treated and untreated samples, between wild-type and knockout backgrounds, or following siRNA-mediated knockdown. The standard workflow involves preparing nuclear extracts under non-denaturing conditions, incubating the extract in the coated wells, washing, and detecting bound c-Jun with the included antibodies. For best stability, store all unopened components refrigerated at 4 °C, protect from light, and limit antibody aliquots to no more than three freeze-thaw cycles. Intended for research use only.
