Description

This Colorimetric Cell-Based ELISA Kit from Assay Genie is designed for the precise detection of phosphorylated C/EBP-epsilon at Thr74 in cell lysates from adherent cells. It is a Cell-Based ELISA reactive with Human samples, with a dynamic range of > 5000 Cells and detection carried out by colorimetric measurement at 450 nm.

The assay is lysate-free and high-throughput, using an indirect ELISA format. Phosphorylated C/EBP-epsilon is captured by phospho-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody and catalyze a colorimetric reaction upon substrate addition. A monoclonal GAPDH antibody is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining step can be used to determine cell density for further normalization between wells.

C/EBP-epsilon is a bZIP transcription factor encoded by the CEBPE gene (Gene ID: 1053, UniProt ID: Q15744, OMIM: 600749), located at chromosomal region 14q11.2 with a molecular weight of 30,603 Da. The protein regulates gene expression linked to cell differentiation, proliferation, and immune response, and phosphorylation at Thr74 modulates its activity. Mutations in CEBPE are associated with Specific Granule Deficiency, a rare congenital disorder. The kit is classified under the Epigenetics and Nuclear Signaling research area and is a phospho-specific reagent.
