Description

From Assay Genie, this Phospho Cell-Based ELISA Kit provides a convenient, lysate-free, high-throughput and sensitive assay for monitoring BTK phosphorylation and expression in cells. The kit enables qualitative determination of BTK (Phospho-Tyr551) concentration through an indirect ELISA format, in which target-specific primary antibodies capture the phosphorylated protein while Dye 1- and Dye 2-conjugated secondary antibodies bound to the Fc region of the primary antibody permit fluorometric detection.

The assay is intended for measuring relative amounts of phosphorylated BTK (Phospho-Tyr551) in cultured cells and for screening the effects that various treatments, inhibitors such as siRNA or chemicals, and activators have on BTK phosphorylation. Two normalisation approaches are provided: a monoclonal antibody specific for human GAPDH serves as an internal positive control for normalising the target RFU values, and an antibody against the non-phosphorylated counterpart of BTK (Phospho-Tyr551) allows RFU values for non-phosphorylated BTK to be used to normalise those for phosphorylated BTK.

Reactivity is reported for Human, Mouse and Rat samples, with a sample type of Cells and a stated dynamic range of > 5000 Cells. Fluorescent plate readings are taken at Ex/Em 651/667 (Dye 1) and 495/521 (Dye 2). Storage is at 4°C for 6 months. The target protein BTK (Bruton tyrosine kinase) is a Tec-family tyrosine kinase; associated identifiers include UniProt ID Q06187 and NCBI Gene ID 695.
