Description

Assay Genie's fluorometric cell-based ELISA kit is designed to monitor phosphorylation and expression profiles of BTK in cells without the need for lysate preparation. The kit supports qualitative measurement of relative levels of phosphorylated BTK in cultured cells, making it suitable for screening the effects of various treatments, inhibitors (such as siRNA or chemicals) and activators on BTK phosphorylation. Detection is achieved through an indirect ELISA format in which target-specific primary antibodies capture BTK, while Dye 1- and Dye 2-conjugated secondary antibodies bind the Fc region of the primary antibody to deliver a fluorometric readout.

The kit is reactive with Human, Mouse and Rat samples and includes a monoclonal GAPDH antibody as an internal positive control for normalising target RFU values, as well as an antibody against the non-phosphorylated counterpart of BTK for further normalisation. The stated dynamic range is > 5000 cells. Storage is at 4 °C with a stability of 6 months. Supplied components include 96-well black cell culture clear-bottom microplates, 10X TBS, quenching buffer, blocking buffer, 15X wash buffer, primary antibody diluent, anti-phospho target antibody, anti-target antibody, anti-GAPDH antibody, dye-1 conjugated anti-rabbit IgG antibody, dye-2 conjugated anti-mouse IgG antibody, and adhesive plate seals.

Database identifiers listed for the target include Gene ID 695, UniProt ID Q06187, OMIM entries 300300/300755/307200 and Unigene Hs.159494. The NCBI gene symbol is BTK and the target is specifically Phospho-BTK (Tyr223). A fluorescent plate reader with two channels at Ex/Em 651/667 and 495/521 is required but not provided.
