Description

From Assay Genie, this Colorimetric Cell-Based ELISA Kit is designed to detect Breast Tumor Kinase (Phospho-Tyr447) in adherent cells with human reactivity. The assay uses an indirect ELISA format with a 450 nm colorimetric readout and has a dynamic range of > 5000 Cells. It is described as a lysate-free, high-throughput solution for detecting Breast Tumor Kinase protein phosphorylation and expression profile in cells, and can be used to measure relative levels of phosphorylated Breast Tumor Kinase in cultured cells as well as to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on Breast Tumor Kinase phosphorylation.

A monoclonal antibody specific for human GAPDH is included as an internal positive control to normalize target absorbance values. Following colorimetric measurement of HRP activity, the Crystal Violet whole-cell staining method may be used to determine cell density, with results analysed by normalizing absorbance values to cell amounts to adjust for plating differences. The target corresponds to PTK6 (UniProt Q13882), a Src-family non-receptor tyrosine kinase selectively expressed in breast tumours and cell lines, where phosphorylation at Tyr447 plays a role in cellular signalling relevant to cancer progression and metastasis.
