Description

A cell-based ELISA kit from Assay Genie designed for the quantitative detection of BRD3 protein in cell lysates and tissue homogenates. The assay uses colorimetric detection measured at 450 nm and is reactive with both human and mouse samples. With a dynamic range of > 5000 Cells, the kit provides a lysate-free, high-throughput workflow for measuring relative BRD3 expression in cultured adherent cells and for screening the effects of treatments, inhibitors, or activators.

The assay follows an indirect ELISA format in which BRD3 is captured by specific primary antibodies and detected through HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. A monoclonal anti-GAPDH antibody is supplied as an internal positive control for normalization, and Crystal Violet whole-cell staining can be used to determine cell density. The kit is suited to research in epigenetics and nuclear signaling, including studies of BRD3 in chromatin binding, transcription regulation, and disease contexts such as cancer and inflammatory disorders. Storage is at 4°C with a stability of 6 Months.

BRD3 (Bromodomain-containing protein 3) binds hyperacetylated chromatin and plays a role in transcription regulation by chromatin remodeling and interaction with transcription factors, including promoting GATA1 binding to its targets and regulating CCND1 transcription. The protein has a reported molecular weight of 60,942 Da. A chromosomal aberration involving BRD3 — translocation t(15;9)(q14;q34) with NUT — produces a BRD3-NUT fusion protein associated with a rare, aggressive midline carcinoma.
