Description

Assay Genie provides a colorimetric, cell-based ELISA for the qualitative detection of phosphorylated BRCA1 at serine 1457 in human samples, suitable for use with adherent cells. The In-Cell ELISA format is lysate-free and high-throughput, allowing researchers to measure relative levels of phosphorylated BRCA1 in cultured cells and to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on BRCA1 phosphorylation.

The assay uses an indirect ELISA format: a target-specific primary antibody captures BRCA1 (Phospho-Ser1457), and an HRP-conjugated secondary antibody binds the Fc region of the primary antibody. Detection is achieved by a colorimetric reaction at 450 nm following substrate addition, with a quoted dynamic range of > 5000 Cells. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalization. After HRP readout, the Crystal Violet whole-cell staining method can be used to determine cell density, allowing absorbance values to be normalized to cell amount.

The target is the BRCA1 gene product (Gene ID: 672, UniProt ID: P38398), a tumor suppressor that plays a central role in DNA repair and the maintenance of genomic stability. Phosphorylation of BRCA1 at serine 1457 has been linked to its function in the DNA damage response and cell cycle regulation, with dysregulation of this event associated with breast and ovarian cancer susceptibility.
