Description

From Assay Genie comes a Colorimetric Cell-Based ELISA Kit designed for the detection and quantification of BRCA1 protein in cultured cells, with adherent cells specified as the compatible sample type. It functions as a lysate-free, high-throughput, and sensitive assay that captures BRCA1 via target-specific primary antibodies, with HRP-conjugated secondary antibodies binding the Fc region of the primary antibody to drive a colorimetric reaction upon substrate addition.

Intended for qualitative determination of BRCA1 concentration, the kit can be used to measure relative BRCA1 levels in cultured cells and to screen for the effects of treatments, inhibitors such as siRNA or chemicals, and activators on BRCA1 expression. Reactivity is Human with a dynamic range of > 5000 Cells. Detection is colorimetric, measured at 450 nm, and the format is a 96-well microplate with recommended storage at 4°C for 6 months. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, while Crystal Violet whole-cell staining may be used to determine cell density, allowing absorbance values to be normalized to cell amounts so plating differences can be adjusted.

BRCA1 is an E3 ubiquitin-protein ligase (UniProt ID: P38398; Gene ID: 672) that plays a central role in DNA repair and acts as a tumor suppressor, with mutations in the gene associated with an increased risk of breast and ovarian cancer. The kit supports research into the molecular mechanisms underlying cancer development and progression, as well as efforts to develop targeted therapies.
