Description

Assay Genie's Bovine P21 (Cyclin-dependent Kinase Inhibitor 1A) ELISA Kit is a Sandwich ELISA in a double-antibody format with colorimetric detection, designed to quantify the cyclin-dependent kinase inhibitor p21 (CDKN1A) in bovine samples. The target acts as a tumour suppressor by inhibiting cyclin-dependent kinases and regulating cell cycle progression, and its dysregulation has been linked to diseases including cancer, making it a valuable biomarker for cell cycle and disease research.

Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit reports a sensitivity of 46.875 pg/mL and a working range of 78.125–5000 pg/mL. Linearity in serum (n=5) is 91-102% at 1:2, 93-102% at 1:4, and 87-105% at 1:8; EDTA plasma (n=5) shows 82-100%, 83-100%, and 84-101% at the same dilutions; heparin plasma (n=5) shows 80-98%, 88-100%, and 80-95%. Recovery averages 96% in serum (range 87-104%), 92% in EDTA plasma (range 85-100%), and 95% in heparin plasma (range 85-105%).

The protocol involves equilibrating reagents to room temperature, a 90-minute primary incubation at 37°C, 60-minute biotin-labelled detection antibody binding at 37°C, 30-minute HRP-Streptavidin (SABC) incubation at 37°C, 10-20 minutes of TMB substrate development in the dark, and absorbance reading at 450 nm after adding the stop solution. The associated Uniprot entry is A5PKM2. Components should be stored at 2-8°C, with the kit stable for 12 months under these conditions.
