Description

This colorimetric sandwich ELISA kit from Assay Genie is designed for the quantitative measurement of Leukemia Inhibitory Factor (LIF) in bovine serum, plasma, cell culture supernatants, cell or tissue lysates, and other liquid samples. LIF is a protein recognised for its ability to inhibit the growth of leukaemia cells and for its roles in promoting stem cell proliferation and differentiation, as well as involvement in immune response, inflammation, and embryonic development.

The double-antibody sandwich format delivers a sensitivity of 18.75 pg/mL with a working range of 31.25-2000 pg/mL. Linearity was demonstrated at 1:2, 1:4 and 1:8 dilutions in serum (88-99%, 85-89%, 80-96%), EDTA plasma (85-105%, 83-96%, 80-96%) and heparin plasma (90-103%, 82-99%, 81-93%). Recovery ranges were 85-102% (average 93%) for serum, 86-99% (average 92%) for EDTA plasma and 93-103% (average 99%) for heparin plasma. Unopened components should be stored at 2-8°C for 12 months.

The assay workflow starts with equilibrating reagents and TMB substrate to room temperature, followed by a primary incubation of standards and samples at 37°C for 90 minutes. A biotin-labelled detection antibody is then added for 60 minutes at 37°C, followed by HRP-Streptavidin (SABC) for 30 minutes at 37°C. Colour is developed with TMB substrate in the dark for 10-20 minutes, the reaction is stopped, and absorbance is read at 450 nm.
