Bovine DAO (Diamine Oxidase) ELISA Kit

From Assay Genie, this Sandwich ELISA kit (Double Antibody format) is designed for the precise quantification of diamine oxidase (DAO) levels in bovine samples, including serum, plasma, tissue homogenates, cell culture supernatant, cell or tissue lysates, and other liquid sample types. Diamine oxidase is an enzyme involved in the metabolism of histamine and other biogenic amines, playing a role in immune response and gut health; dysregulation of DAO activity has been linked to conditions such as histamine intolerance and inflammatory bowel diseases, making it a valuable biomarker for related research.

The assay uses a colorimetric detection method with a sensitivity of 0.469 ng/mL and a working range of 0.781–50 ng/mL. The kit is supplied with components for a standard sandwich ELISA workflow, including a pre-coated dismountable microplate, lyophilized standard, biotin-labeled detection antibody, HRP-Streptavidin conjugate (SABC), TMB substrate, sample and antibody dilution buffers, SABC dilution buffer, stop solution, 25X wash buffer, plate sealer, and a technical manual. The TMB-based procedure includes incubation at 37°C for antigen binding, detection antibody binding, HRP-Streptavidin binding, and color development, with absorbance read at 450 nm after stop solution is added. Storage is at 2–8°C for 12 months.

Validated linearity and recovery values are provided for serum, EDTA plasma, and heparin plasma. Linearity across 1:2, 1:4, and 1:8 dilutions is 88–98%, 88–105%, and 92–101% for serum; 85–93%, 85–102%, and 82–85% for EDTA plasma; and 83–88%, 81–100%, and 85–98% for heparin plasma. Recovery ranges are 93–105% (average 98%) for serum, 86–103% (average 93%) for EDTA plasma, and 88–101% (average 95%) for heparin plasma.
