The Assay Genie bovine CD40L ELISA kit enables quantitative measurement of Cluster of Differentiation 40 Ligand in cattle-derived research specimens through an immunoassay workflow that produces a colorimetric signal on a standard microplate absorbance reader. CD40L, also known as CD154 or TNFSF5, is a type II transmembrane protein of the TNF superfamily that serves as the principal ligand for CD40 on B cells, dendritic cells, and macrophages, driving B-cell proliferation, germinal center formation, immunoglobulin isotype switching, and antigen-presenting cell activation.

Each kit is configured for 96 determinations and is supplied with a pre-coated microplate, biotin-conjugated detection antibody, streptavidin-HRP reagent, calibrator standards, wash buffer, substrate buffer, and stop solution. The sequential add-incubate-wash protocol relies on a TMB substrate that develops a measurable color change upon reaction termination, captured as absorbance values on a standard plate reader at the assay's recommended wavelength.

Common research applications include quantifying soluble CD40L release from concanavalin-A or pokeweed mitogen-stimulated bovine peripheral blood mononuclear cell cultures, tracking plasma CD40L dynamics in Mycobacterium bovis or BVDV challenge studies, profiling vaccine-elicited immune responses in cattle herds, and assaying lymph node and mucosal tissue lysates. The kit is intended for research use only and is not validated for clinical diagnostic or therapeutic decision-making.
