Description

Assay Genie provides a Sandwich ELISA, Double Antibody format kit for the quantitative measurement of CD46 in bovine samples. Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay reports a sensitivity of 9.375 pg/mL and a detection range of 15.625–1000 pg/mL, with storage at 2–8°C for 12 months.

Linearity testing (n=5) in serum showed 87–103% at 1:2, 85–102% at 1:4, and 92–105% at 1:8 dilution. EDTA plasma ranged 86–98%, 85–99%, and 82–100% across the same dilutions, and heparin plasma showed 87–98%, 86–98%, and 85–99%. Recovery averaged 96% for serum (range 90–102%), 97% for EDTA plasma (range 89–100%), and 95% for heparin plasma (range 91–103%).

CD46, also referred to as MCP or Membrane Cofactor Protein (Uniprot Q6VE48), is described as a key protein involved in immune response regulation and essential for protecting cells from complement-mediated damage. Dysregulation of CD46 has been implicated in autoimmune diseases and inflammatory disorders, making it a valuable biomarker in bovine disease models. The procedure includes 90 minutes of primary incubation at 37°C, 60 minutes with biotin-labeled detection antibody, 30 minutes with HRP-streptavidin, TMB color development in the dark for 10–20 minutes, and absorbance measurement at 450 nm after adding stop solution.
