Caspase-4 (CASP4) is an inflammatory cysteine protease that mediates the non-canonical inflammasome pathway, cleaving gasdermin D to drive pyroptosis and the release of pro-inflammatory cytokines such as IL-1β and IL-18. In bovine research, CASP4 levels are commonly examined in studies of mastitis, respiratory infections, and systemic inflammatory responses, supporting veterinary and livestock immunology investigations.

This Assay Genie sandwich ELISA is supplied as a 96-well microplate kit with a colorimetric detection method, typically read at 450 nm on a standard plate reader. The kit has been validated for bovine serum and plasma samples, and components should be stored at 2–8 °C; it is intended for research use only and is not approved for diagnostic procedures.

Each Assay Genie sandwich ELISA kit is shipped with a pre-coated 96-well microplate, lyophilized standards, detection antibody, streptavidin-HRP conjugate, wash buffer, substrate solution, and stop buffer, sufficient for one full plate run. The standard protocol typically requires 100 µL of sample per well, with a total assay time of approximately 3–4 hours depending on incubation steps. Researchers should bring all reagents to room temperature before use and avoid repeated freeze-thaw cycles of reconstituted standards and antibodies. The assay principle relies on a pair of antibodies recognizing distinct epitopes on bovine CASP4, with the captured analyte detected via a colorimetric TMB substrate read at 450 nm on a standard microplate reader.

Typical applications include quantitative measurement of CASP4 in bovine serum and plasma samples from studies of inflammatory disease, mastitis, and pulmonary infection models, as well as investigation of the non-canonical inflammasome pathway and pyroptosis-related cytokines. The kit is intended for research use only and should not be used for clinical diagnostic or therapeutic decision-making.
